In this video, Prof. Markus Eckstein of the Institute of Pathology at Erlangen University Hospital discusses the clinical significance of PTEN alterations in metastatic hormone-sensitive prostate cancer (mHSPC). The loss of the tumor suppressor PTEN permanently activates the PI3K/AKT signaling pathway and promotes a more aggressive course of the disease; this affects about one-quarter of patients with newly diagnosed mHSPC.
FISH and NGS are available for testing to identify genomic alterations, as is immunohistochemistry for the direct detection of the PTEN protein. Against this backdrop, there is a significant unmet medical need and, at the same time, a promising avenue for targeted therapies, which requires reliable determination of PTEN status in clinical practice.
