Darolutamide + ADT for Metastatic Hormone-Sensitive Prostate Cancer: ARASEC

For metastatic hormone-sensitive prostate cancer (mHSPC), the combination of an androgen receptor pathway inhibitor (ARPI) and androgen deprivation therapy (ADT) has become established internationally as the guideline-recommended standard of care, to the extent that ADT monotherapy is now rarely used as a control arm in clinical trials, particularly in the United States. To nevertheless generate additional evidence for the combination of darolutamide and ADT specifically in U.S. patients, the ARASEC study was designed in parallel with the global ARANOTE study. ARASEC compares darolutamide + ADT with an external ADT monotherapy comparison arm from the Phase 3 CHAARTED trial, selected using a recognized propensity score matching method.

The course will cover the study’s innovative design, the matching methodology, and the efficacy, safety, and sensitivity analyses of the study, as well as how these findings fit into the current evidence base.
