Acute pain is a major clinical challenge and is often treated with opioids, despite the risk of abuse and dependence. The voltage-gated sodium channel NaV1.8 represents a promising therapeutic target for pain treatment, as it plays a crucial role in the transmission of nociceptive signals and is selectively present in peripheral nociceptive neurons of the dorsal root ganglia. Today's two phase 2 clinical trials are therefore analyzing the efficacy and safety of VX-548 - an oral, highly selective NaV1.8 inhibitor - in patients with acute postoperative pain following abdominoplasty or bunionectomy.
